My research is focused on the genetics and epigenetics of alcoholism including fetal alcohol spectrum disorders (FASD). In recent years, my research team have taken on the Neuroepigenetics mechanismof FASD by studying intrinsic DNA methylation program during brain development in response to prenatal alcohol exposure. We are analyzing neural development and defects at molecular and chromatic levels using high throughput analysis of DNA methylation and histone modification. We recently elucidated that an epigenetic program, which includes 5-methylcytosine (5mC), 5-hydroxylmethylcytosine (5hmC) with distinctive roles, progresses during development of hippocampus, cortex, and during adult neurogenesis. This program is found to be affected by prenatal alcohol exposure at epigenomic level, and the differentially methylated regions (DMRs) can be identified at CpGs at single gene levels with methyl-sequencing and pyrosequencing. Recently, we have taken on epigenetic editing to repair the erratic DNA methylation at specific DMRs to analyze their roles on transcription and actions on phenotype.
The second focus of my current research is to investigate how familial drinking preference are carried across generations through genetic and reinforced by epigenetic mechanisms using high and low drinking rat model HAD/LAD line and P/NP line. Using next generation sequencing, the genomic SNP signature of duplicate HAD/LAD lines revealed that genetic background of high alcohol drinking behavior exhibits a polygenic architecture consistent with a complex trait. A large number of genes were identified, each with a small effect. The landscape of the genetic signature indicated that environmentally influenced variation is mainly due to regulatory elements. We are currently investigating the second arm of the genetics of alcoholism by looking into DMRs through inheritance and alcohol-induction using methyl-sequencing analysis.
I have directed various grant projects e. g. involve “Epigenetics of fetal alcohol syndrome”, and “Genomics of Alcohol preference”. I have also served as President and advisor for the FASDSG (FASD Study group, USA), and currently an advisor in the Consortium on the Neurobiology of Adolescent Drinking in Adulthood (NADIA).
